In this study, we investigated the role of the extracellular matrix (ECM) in the underlying biology of lung adenocarcinoma using an autochthonous mouse model that recapitulates the complexity of cancer initiation and progression to characterize the ECM composition of normal lung, fibrotic lung, lung tumors, and metastases. We isolated normal, healthy lung tissues from wild-type (WT) mice and microdissected KrasG12D/p53-/- primary lung tumors (KP tumors) and associated metastases to the mediastinal lymph node. Quantitative mass spectrometric profiling of the ECM composition of normal lung, fibrotic lung (from bleomycin-treated mice), primary lung tumors, and lung metastases to the lymph nodes uncovered specific signatures distinguishing these tissues. This gene set lists the matrisome proteins detected in significantly different abundance in lymph node metastasis arsing from KP primary tumors as compared to normal lung.
