Full description or abstract In this study, we aimed to characterize changes in the extracellular matrix (ECM) composition during insulinoma progression using a quantitative proteomics approach. To do so, we chose a mouse model of insulinoma, a subtype of pancreatic neuroendocrine tumors, characterized by the expression of SV40 large T antigen (Tag) in pancreatic beta-cells driven by the rat insulin promoter (RIP) (RIP1-Tag2 model). This model follows a well-defined timeline of tumor progression: hyperplastic islets appear by 4 weeks of age, angiogenic islets by 7-9 weeks, solid tumors at 10 weeks, and large and invasive adenomas by 12-13 weeks. Using quantitative proteomics based on isobaric peptide labeling (iTRAQ), we profiled the ECM proteomes or matrisomes of various stages: normal pancreatic islets, hyperplastic islets, angiogenic pancreatic islets, and insulinomas. We focused on ECM and ECM-associated proteins along with proteins found in only one of the two replicates but with at least two unique peptides. Applying a moderated F-test, we identified proteins detected in statistically significantly different abundance in tumor samples (hyperplastic, angiogenic islets or isulinomas) as compared to normal pancreatic islets. The gene set lists the matrisome proteins detected in significantly higher abundance during the hyperplastic stage of insulinoma progression as compared to normal islets and later stage insulinomas.
